WSG, a water soluble glucose-rich polysaccharide, is isolated from fucose-containing polysaccharide fraction F3 of Ganoderma lucidum. However, less study reported the effects of WSG on solid tumor. Herein, we examined the effects of WSG on two types of cancer cells including lung cancer and oral cancer. Specifically, we focused on dissecting the mechanism of WSG-suppressed lung and oral cancer cells and further analyze the efficacy of WSG combined with clinical chemotherapeutic drug, cisplatin on cancer treatment. We found that WSG suppressed cell viability and colony formation in various lung cancer A549 and LLC1 cells as well as oral cancer SAS and HSC3 cells in vitro. In addition, WSG inhibited cell mobility of lung cancer cells. Functional studies revealed that WSG reduced phosphorylation of epidermal growth factor receptor (EGFR) and AKT, which are correlated with proliferation of lung and oral cancer cells. WSG reduced EGFR and Transforming growth factor beta receptor (TGFβR) in lung cancer cells. Moreover, WSG significantly potentiated cisplatin-induced cytotoxicity via enhancement of apoptotic responses in lung and oral cancer cells. Meanwhile, WSG effectively inhibited tumor growth in lung cancer cells-bearing mouse. In conclusion, WSG may be a potential supplement for cancer therapy.
